On Monday, we did tail measurement for lymphedema mice model (week 4 after surgery). I also shadowed the fixation and preservation of mice intestines, lymph nodes, and colons for wild type and PI3K ko mice. Subsequently, we did RNA isolation for PCR. The goal of PCR is to compare the difference in baseline expression of TP in HUVEC and LEC. Cells were harvested from 6-well plate and separated into RNA, protein, and organic phases using Trizol method. After contrifugation, isopropanol were used to condense nucleic acid from the supernatant.

On Tuesday, we prepared for flow cytometry (scheduled on Wednesday) by collecting cells from mice tail skin. We were interested in quantifying the T cells lymphocytes, fibroblasts, macrophages, mast cells, and LEC populations. I also shadowed the PCR.

On Wednesday, we performed the flow cytometry for mice skin cells. The cells were stained for fibroblasts, macrophages, mast cells, LEC, and T cells using specific antibodies with fluorophores.
