Harry Zou - Week 5

 This week was a short week with the 4th of July holiday. I spent this week focused on my research project and preparing the gDNA samples from last week to be sent out for sequencing. I first cleaned up the PCR product from last week which included 3 different samples at a gradient of 4 different annealing temperatures. Then, I prepared the various buffers needed to run a DNA gel and then ran the cleaned-up PCR products through the gel. For all of the amplification conditions, I observed a band at 3500 bp which is the size of the gene I am trying to sequence. However, only 68 degrees had specific bands so moving forward I will be running PCR with a 68 degree annealing step.
